Drugs inducing T-cell mediated cutaneous adverse reactions and contact sensitizers evoke similar responses in THP-1 cells

چکیده
Background Contact sensitizers induce an innate immune response in dendritic cells (DC) enhancing antigen presentation and T cell response. Little is known concerning the effect of systemic drugs causing T-cell mediated cutaneous adverse drug reactions (CARDs). Therefore, we studied the in vitro effect of drugs on THP-1 cells, a monocyte cell line widely used to show activation by contact sensitizers.

Cancer Therapy: Clinical Cytotoxic Cutaneous Adverse Drug Reactions during Anti-PD-1 Therapy
Purpose: Immunotherapy has experienced impressive progress in cancer treatment. Antibodies against PD-1 improved survival in different types of cancer including melanoma. They are generally well tolerated. However, skin toxicities including pruritus, rashes, and vitiligo are reported. Although frequent, they have not been characterized further yet. In this analysis, we aimed to systematically a...
متن کاملHMOX1 and NQO1 genes are upregulated in response to contact sensitizers in dendritic cells and THP-1 cell line: role of the Keap1/Nrf2 pathway.
Electrophilicity is one of the most common features of skin contact sensitizers and is necessary for protein haptenation. The Keap1 (Kelch-like ECH-associated protein 1)/Nrf2 -signaling pathway is dedicated to the detection of electrophilic stress in cells leading to the upregulation of genes involved in protection or neutralization of chemical reactive species. Signals provided by chemical str...

Contact hypersensitivity (CH) to simple haptens and acute allograft rejection are generally regarded as T cell-mediated responses. However, at the present time, considerable controversy exists concerning the precise phenotype of the effector cells that are the proximate causes of inf lammation and tissue destruction. In transplantation immunity, the assumption of a decade a g o t h a t cytotoxi...
متن کاملPatch testing in the investigation of non-immediate cutaneous adverse drug reactions to metamizole.
Metamizole [INN; dipyrone (BAN, USAN)] is a pyrazolone derivative and one of the most commonly prescribed analgesics worldwide (1). Along with non-steroidal anti-inflammatory drugs (NSAIDs), metamizole is very frequently involved in hypersensitivity reactions (2). Immunologically mediated hypersensitivity reactions to metamizole can be immediate (IgE-mediated) or non-immediate (T cell-mediated)...
متن کاملEffects of berberine on the secretion of cytokines and expression of genes involved in cell cycle regulation in THP-1 monocytic cell line
Objective(s): Current acute myeloid leukemia (AML) therapeutic strategies have irreversible side-effects. Berberine (BBR) is an isoquinoline alkaloid, which has been known as an aryl hydrocarbon receptor (AhR) ligand. AhR is a cytoplasmic receptor, which is involved in the regulation of cellular and immune responses. Here, we investigated the expression profile of genes involved in the cell cyc...
